The pharmaceutical grade, natural SAM-e in Jarrow Formulas’ SAM-e Joint is made naturally by microbiological fermentation in a salt form that ensures a stable, 2+year shelf life of SAM-e, without compromising chemical structure or biological activity (Univ. of MD Medical Center 2004). In addition, it is manufactured under low temperatures and humidity, and then enteric-coated and sealed into blister packs, to ensure a biologically active product.

SAM-e And Glucosamine: a Joint effort
Found in every living cell, SAM-e is known as a universal methyl donor, providing methyl groups for numerous reactions in the body, including many that are critical for healthy cartilage and central nervous system function.
A metabolite of the amino acid methionine, SAM-e is a chiral molecule which has two forms: a biologically active form, the (S,S) structure, and the(R,S) structure which is biologically inactive. Jarrow Formulas’ SAM-e contains 68- 80% of the (S,S) form of SAM-e, in the highest active level available. Glucosamine HCl, a naturally occurring amino glycan (or amino sugar), is the fundamental building block for making glycolipids, glycoproteins, hyaluronate, and proteoglycans. It is component of joint and intestinal tissue, and involved in the production of synovial fluid that lubricates the joints. It is synthesized by transamidase enzymes from glucose and an amine group from glutamine and stabilized with HCl (hydrochloric acid). Glucosamine undergoes sulfation by SAM-e metabolites to optimally support healthy joints and connective tissue.

Joint Strength
SAM-e works in multiple ways to support healthy joints and connective tissue. Clinical trials have shown improvements in joint discomfort and stiffness, while in vitro and animal studies have shown SAMe to stimulate production of new cartilage.
In a randomized double-blind trial comparing SAMe and a conventional treatment, 56 subjects were given either 1200mg/day of SAMe or 200mg/day of the conventional treatment for 8 weeks and assessed for discomfort, functional health, and mood.
After 8 weeks, both groups showed significant improvements in all parameters compared to baseline, with no significant difference between the treatment groups.
SAM-e also supports healthy connective tissue through its role in trans-sulfuration.
In an animal study, 24 rabbits were given 30mg or 60mg/kg/day of SAM-e or placebo control for 12 weeks; they found the number of cells, the depth of the cartilage and level of proteoglycan synthesis were all significantly increased in both SAM-e treatment groups, compared to controls.
Researchers speculate that SAM-e may stimulate the incorporation of sulfate into proteoglycans and enhance joint tissue proliferation.
Numerous meta-analyses have confirmed the efficacy of glucosamine and strong randomized, placebo-controlled studies, including the well-known GAIT (Glucosamine/Chondroitin Arthritis Intervention Trial) trial, have shown its efficacy in improving joint comfort.
In a double-blind trial of 178 patients with knee discomfort, 1500 mg of glucosamine sulfate led to significant improvement in symptoms after 4 weeks and continued to improve 2 weeks after treatment was stopped.
In addition, a 2013 study of over 11,000 adults showed a significant improvement in joint comfort, functional mobility and use, and quality of life with glucosamine.

Liver Detoxification & Brain/Cognitive Health
SAM-e also works in multiple ways to support the liver’s detox pathways, both as donor for methylation reactions, a participant in the synthesis of glutathione, and as a key metabolite that regulates hepatocyte growth.
A randomized, double-blind, placebo-controlled clinical trial of 123 liver patients treated with either 1200mg/day of SAM-e or placebo for 2 years found those given SAM-e had significantly improved longevity and significantly enhanced liver function over controls.
SAM-e’s role in methylation is also responsible for its broad effect on brain and cognitive health.
As a methyl donor, SAM-e plays a key role in the synthesis of neurotransmitters and phosphatidylcholine; it is required for the catecholamine pathway to function properly, and regulated by a feedback system. Thus, SAM-e plays a significant role in ensuring normal pathway function.
